Lack of therapeutic options for patients with metastatic osteosarcoma has led to an investigation into novel therapeutic approaches including oncogene-targeted therapies. In this study, we have used two small interfering RNAs (siRNAs) targeted to ERBB2 to investigate the therapeutic potential of this approach in osteosarcoma cells. siRNAs were transfected into the osteosarcoma cell lines MG-63 and HOS, which express the oncogenic form of ERBB2 due to chromosomal translocation. Down-regulation of ERBB2 was confirmed by quantitative RT-PCR and Western blot analysis. Cell proliferation, apoptosis, migration, and invasion were evaluated by cell proliferation, TUNEL and annexin V/propidium iodide (PI) staining, wound healing, and Matrigel invasion assays, respectively. Proliferation was significantly decreased in both MG-63 and HOS cells transfected with siERBB2. Similar results were obtained using the Matrigel invasion assay. siERBB2-transfected cells exhibited increased apoptosis as determined by Annexin V/PI staining.
